If I build a three floors house with roof and walls, and I put an activated Heating or Cooling Coils on the first floor, will whole house be affected?

If there's a stair case or other empty space leading up to the other floors, yes, it will reach it. The range of the coils is generally large enough for any house. If not, just place another coil near the top.
